PERIPHERAL TOLERANCE. Although central-tolerance mechanisms are efficient, they cannot eliminate all self-reactive lymphocytes, in part because not all self-antigens are expressed at the primary site of lymphocyte development—the thymus. Peripheral tolerance is the second branch of immunological tolerance, after central tolerance. It takes place in the immune periphery (after T and B cells egress from primary lymphoid organs ). Its main purpose is to ensure that self-reactive T and B cells which escaped central tolerance do not cause autoimmune disease . Central tolerance prevents the maturation and egress of autoreactive immune cells, for example via clonal deletion of T cells in the thymus 1.
